The reserves' unbeaten start to the season ended on Saturday when they were beaten 2-1 at Hebburn Town Reserves in the Alan Hood Cup.
Quakers had won their first two league games, but that winning form didn't transfer itself to the Cup.
They were two goals down at half time, but despite a Lewis Walton goal in the second half, they couldn't turn the game around.
Joint manager Mark Fanning said; "We gifted them two goals in the first 20 minutes. I thought we outplayed them after that, had loads of possession, but we could only score one. Apart from the mistakes, I thought our performance was good.
"It was our first defeat of the season, we'll see now how our players react to it now."
The reserves next game is at Sunderland West End on Wednesday night.
There were no first team players in the squad, as Kevin Burgess, Tom Portas, Jordan Richards and Joe Wheatley, who had played at Stokesley last Wednesday, were all with the first team for the home win over Alfreton.
